After colonizing your grain spawn (e.g., rye berries, millet, or brown rice), mix it thoroughly with a pasteurized bulk substrate and layer it evenly across the base of the monotub to a depth of about 3–4 inches. If using a casing layer (especially for species like Psilocybe cubensis or Pan. cyanescens), apply it evenly on top of the substrate.
Place the lid on and tape over or plug the ventilation holes during the initial colonization phase to retain CO₂ and humidity. Once full colonization occurs (usually 10–14 days), remove the tape or hole plugs to allow filtered fresh air exchange through the gasketed ports. Mist the sidewalls of the tub lightly with distilled water once or twice daily to maintain ambient humidity of 85–95%—avoid direct misting of the mycelium.
The monotub should be placed in a well-lit room with indirect natural light or a 6500K full-spectrum LED source for 12 hours daily. Maintain room temperature between 70–75°F (21–24°C). Within 5–10 days of initiating fruiting conditions, you should see pin formation, followed by maturing fruiting bodies ready for harvest within a week.
