Instructions for Use
Understanding the Culture Slant
A culture slant consists of nutrient-rich agar inoculated with healthy mushroom mycelium. It is designed for propagation and long-term storage.
Propagation from the Reishi Culture Slant
- Prepare a Sterile Work Environment:
- Use a laminar flow hood or glove box to ensure a clean environment.
- Sterilize tools such as scalpels, tweezers, and containers with flame or alcohol.
- Inoculating Agar Plates:
- Carefully open the culture slant, sterilize the tube mouth with a flame, and avoid contamination.
- Use a sterile scalpel to scrape a small amount of mycelium from the agar.
- Transfer the mycelium to sterilized agar plates and seal them.
- Store plates at 75–80°F (24–27°C) for colonization.
- Preparing Grain Spawn:
- Once the agar plates are colonized, cut small sections of the colonized agar.
- Place the mycelium-covered agar pieces into sterilized grain jars (rye, millet, or oats).
- Allow the grain to fully colonize at room temperature, shaking the jars periodically to spread growth evenly.
- Substrate Transfer:
- Once the grain spawn is fully colonized, transfer it to hardwood substrates, such as sawdust, hardwood chips, or logs.
- Reishi thrives on hardwood-based substrates like oak, maple, or beech.
- Fruiting Conditions:
- Expose the substrate to the following fruiting conditions:
- Temperature: 70–80°F (21–27°C).
- Humidity: 85–95%.
- Light: Low-intensity indirect light (12 hours per day).
- Fruiting begins in 4–6 weeks, resulting in the iconic shiny, red-brown conks of the Reishi mushroom.

- Short-Term Storage: Store the culture slant at room temperature for up to 1–2 weeks.
- Long-Term Storage: Refrigerate the culture slant at 35–40°F (2–4°C) to extend viability for up to 6–12 months.
- Sterile Handling: Always work with sterile tools and in a clean environment to prevent contamination.
