Understanding the Culture Slant
A culture slant is a test tube containing nutrient-rich agar inoculated with healthy mycelium. It is designed for long-term storage and mycelial propagation.
Propagation from the King Oyster Culture Slant
- Prepare a Sterile Work Environment:
- Work in a clean, sterile area using a glove box or laminar flow hood.
- Sterilize tools such as scalpels, tweezers, and jars with alcohol or flame.
- Inoculating Agar Plates:
- Open the culture slant carefully and flame the mouth of the tube to sterilize it.

- Hardwood sawdust
- Supplemented sawdust blocks (bran or gypsum)
- Straw

- Fruiting Conditions:
- Introduce the following fruiting conditions:
- Temperature: 60–70°F (15–21°C).
- Humidity: 85–95%.
- Light: Indirect light for 12 hours per day.
- Fruiting typically occurs within 10–14 days, producing large clusters of thick, meaty King Oyster mushrooms.
